JJC8-088 is a potent dopamine transporter (DAT) inhibitor derived from the wakefulness-promoting agent modafinil. With a substantially higher affinity for the dopamine transporter compared to modafinil, JJC8-088 exhibits cocaine-like psychostimulant effects and robustly increases dopamine levels in the nucleus accumbens.
Unlike modafinil, which is an atypical dopamine reuptake inhibitor, JJC8-088 acts as a typical cocaine-like dopamine reuptake inhibitor, stabilizing the DAT in an outward-facing open conformation.
JJC8-088 has been shown to effectively reduce compulsive self-administration of methamphetamine in rats and increase extracellular dopamine levels in rats with a history of cocaine self-administration.
