Citicoline, also known as cytidine diphosphate-choline (CDP-choline) or cytidine 5′-diphosphocholine, is an intermediate in the biosynthesis of phosphatidylcholine, a major component of cell membranes. It is naturally occurring in human and animal tissues, particularly in organs. Citicoline is available as a dietary supplement in over 70 countries under various brand names, including CereBleu, Cebroton, Ceraxon, and Cognizin. When taken as a supplement, citicoline is hydrolyzed into choline and cytidine in the intestine, which then cross the blood-brain barrier and are reformed into citicoline by the enzyme CTP-phosphocholine cytidylyltransferase.
Citicoline has been studied for its potential benefits in cognitive impairments, ischemic stroke, and vision protection, particularly in glaucoma patients. It may have neuroprotective effects due to its preservation of cardiolipin and sphingomyelin, restoration of phosphatidylcholine levels, and stimulation of glutathione synthesis. Citicoline is water-soluble with high oral bioavailability, and its plasma levels peak one hour after ingestion. It is excreted primarily as CO2 in respiration, with the remainder excreted in urine.
