Esmodafinil, also known as (S)-modafinil or (+)-modafinil, is the enantiopure (S)-(+)-enantiomer of modafinil.
Unlike armodafinil ((R)-(−)-modafinil), esmodafinil has never been marketed independently. It is suspected to be less clinically effective for conditions like narcolepsy, shift work sleep disorder, and obstructive sleep apnea, which modafinil and armodafinil treat.
Esmodafinil has a 3-fold lower affinity for the dopamine transporter (DAT) compared to armodafinil and a shorter elimination half-life (3–5 hours vs. 10–17 hours). It has been researched for cocaine addiction treatment but is less favored due to its short half-life.
