SKF-38393 is a synthetic compound belonging to the benzazepine chemical class, known for its role as a selective D1/D5 receptor partial agonist. This compound exhibits stimulant and anorectic properties, making it a subject of interest in various research fields. Studies have shown that SKF-38393 can enhance temporal order memory performance in maternally deprived rats and reverse cocaine-conditioned place preference in mice. Additionally, it has been found to induce GAP-43 expression and long-term potentiation in the hippocampus in vivo.
In a study published in Neuroscience, researchers investigated the effect of SKF-38393 on the spontaneous release of glutamate from cultured rat hippocampal neurons. The results demonstrated that SKF-38393 increased the frequency, but not the amplitude, of tetrodotoxin-resistant excitatory postsynaptic currents, suggesting a presynaptic site of action. This effect was blocked by the D1-dopaminergic receptor antagonist SCH-23390 and protein kinase A inhibitors, indicating a mechanism dependent on D1-type dopamine receptors and protein kinase A.
