Apimostinel is an investigational antidepressant under development by Gate Neurosciences for the acute treatment of major depressive disorder (MDD). It acts as a novel and selective modulator of the NMDA receptor, enhancing synaptic plasticity without the psychotomimetic effects associated with other NMDA receptor antagonists like ketamine. Apimostinel is an orally active, potent partial agonist of the glycine site on the NMDA receptor, with a mechanism of action similar to rapastinel (GLYX-13) but with 1000-fold greater potency in vitro.
