ISRIB (Integrated Stress Response Inhibitor) is an experimental drug that reverses the effects of eIF2α phosphorylation.

Discovered in the laboratory of Peter Walter at UCSF, ISRIB inhibits stress granule formation induced by eIF2α phosphorylation. It readily crosses the blood-brain barrier, with a half-life of eight hours, and has shown significant nootropic effects in mice, enhancing spatial and fear-associated learning.

ISRIB has demonstrated potential in reversing cognitive impairments from traumatic brain injury, improving memory in aged mice, and treating neurodegenerative diseases like Alzheimer’s, Parkinson’s, and ALS. Its mechanism involves accelerating the assembly of eIF2B into an active form, counteracting the effects of eIF2α phosphorylation.
